Dhawan kept Mumbai bowlers at bay and reached his half-ton in 36 balls. The opener hit his fifth boundary to complete his half-ton and guided his side cross 100 in the 12th over.
His knock was studded with six boundaries and two magnificent sixes.
Earlier, pacer Lasith Malinga gave Mumbai Indians first success by removing Parthiv Patel after a flying start.
Patel slammed 14-ball-26 and shared 38-run stand with Dhawan before being caught by Rayudu off Malinga in the fourth over.
Sunrisers Hyderabad won the toss and elected to bat.
Mumbai replaced Maxwell, Abu Nechim with Pollard, Dhawal. Hyderabad remained unchanged.
