Jayawardene and Tharanga kept Indian attack at bay for as long as 39th over before spinner Ravichandran Ashwin gave the world champions their first breakthrough in 38.4 overs.
Soon after Jayawardene's wicket, his opening partner Tharanga completed his 13th ODI century off 124 balls, which included eleven hits to the fence.
After hitting his 16th ODI century, Jayawardene tried to accelerate the Lankan innings but failed to clear the field while trying to reverse sweep Ashwin. Mahela smashed two sixes and nine boundaries in his quick-fire 107 off 112 balls.
India bowlers were seen struggling for a breakthrough as Lankan openers guided the side past 200 in 37.2 overs.
Jayawardene reached his fifty in 60 balls with the help of four boundaries and a six while Tharanga scored his fifty off 76 deliveries. Tharanga hit five boundaries.
Sri Lanka's first fifty runs came off 65 balls. However, the openers accelerated the scoring rate after the first powerplay.
Earlier, India's replacement skipper Virat Kohli won the toss and decided to field against the Islanders.
Kohli said it was a great honour for him to lead the Indian side. He was appointed skipper after MS Dhoni was ruled out due to injury.
India replaced Dhoni and Bhuvneshwar Kumar with Murali Vijay and Shami Ahmed.
Sri Lanka, on the other hand, dropped Jeevan Mendis and Ajantha Mendis and brought in Kusal Perera and Senanayake.
